Follow these phases in order to protect the missable work and keep the Dead Cells Platinum cleanup efficient.
Phase 1: Learn the game and unlock the runes
Play freely. Nothing is missable, death is the core mechanic, and there is no multiplayer.
Invest cells in the forge as you go. Better equipment on each run makes everything later considerably easier.
Acquire all runes, since they gate access to the 17 biomes and 20 trophies depend on reaching them.
Do not unlock the Cursed Sword yet. Leaving it locked keeps that trophy off RNG until you are ready.
Beat each of the four bosses for the first time as you progress.
Phase 2: The Boss Stem Cell progression
Work up to finishing the game with four Boss Stem Cells active. Only four are required, not five.
Each step up is a meaningful jump, but if you reached this point you are capable of the next one.
The fifth Boss Stem Cell is a later addition and is optional, so stop at four unless you want it for its own sake.
Doing this before the weapon-restricted runs is deliberate. After four Stem Cells, playing with none is a cakewalk.
Phase 3: The four no-damage boss kills
Concierge, Conjunctivius, Time Keeper and Hand of the King, each without taking a single hit. This is the platinum.
Remember Concierge and Conjunctivius cannot appear in the same run, so budget separate attempts for each.
For the Time Keeper, you can parry her charged attack with a shield if you time it right, including when caught in her chains.

Phase 4: The restricted runs and the platinum
Finish the game with the starter sword, bow or shield, and complete the Cursed Sword run.
Unlock the Cursed Sword now, then play through with your preferred weapons and pick it up just before the final boss.
Clear the remaining oddities, the elevator kill, the Challenge Rift and the secret zones.
With four Stem Cells behind you, these runs are comfortable rather than punishing.

Do Not Unlock the Cursed Sword Yet
One trophy requires finishing the game with the Cursed Sword. If you leave it locked until you are ready, you can play through with your preferred weapons and pick the sword up just before the final boss, rather than relying on RNG to hand it to you at the wrong moment. This is a decision made many hours before the trophy matters.
Efficient route
The Four No-Damage Boss Kills
, an H3 each. Concierge, Conjunctivius, Time Keeper and Hand of the King, each beaten without taking a single hit. These are the whole difficulty of the platinum. Include the Time Keeper parry tip, her charged attack can be parried with a shield if timed correctly, even when you are caught in her chains.
Order Matters, Do the Hard Runs First
After beating the game with four Boss Stem Cells active, playing with none is straightforward. So clear the Stem Cell progression before the easier weapon-restricted runs rather than after.
